What is the role of the internal control or housekeeping gene in real time PCR?
The housekeeping gene in the RT-PCR is a general gene present in the target DNA under test. It is used to confirm that the RT-PCR process is working well. For example if we use RT-PCR for detection of specific mutation in human DNA. If we use the primers of this mutation only and the DNA under test not contain this mutation the result of pcr will be negative but we may not sure whether the mutation is absent or there is a problem in the work procedure gave us a false negative?
To be sure we have to use two types of primers, first the specific primer for the mutation, and the second for a general gen in the human gene (in this example) which is called as housekeeping gene. If the housekeeping gene curve will be exhibited, this will indicate that the RT-PCR process is successfully performed so that the absence of the mutation is confirmed (Aboubakr, 2012).
